Daily Connectivity Loss

Right now at the school I am in the middle of MAP Testing (A standardized, required test).  The software for the testing uses a shared drive on the server to store the databases of student information and question banks.  The accounts that log into the system are setup to use the home drive as this shared folder.  This particular configuration has worked flawlessly for over 5 years now.  The last five days have been an absolute nightmare!  Every day at 2:12pm CST every computer connected into the MAP system suddenly halts all activity in the MAP system and returns a generic error about connection loss to the shared folder.  Tech support has been of no help to me and has pretty much said that they can do nothing else for me.  

I have checked everything from system logs on workstations to logs on the server and everything else.  The server is using WSUS as its method of updating which has been suspended for the duration of the testing session.  There are no AV scans taking place and no other activities happening on this server that I can find.  

The server is running Windows 2008 and is running as a client on a ESXi 5.5 host.  This is the only vm client running on this particular host at this time; all other vm clients have been migrated to another host.  

I don't know what is causing this or where or how to even start looking for it.
